Methylation, which contributesto epigenetic inheritance, can occur either through DNA methylation or protein methylation. DNA methylation in vertebrates typically occurs at CpG sites, which results in the conversion of the cytosine to 5-methylcytosine. 1-2% of the human genome has CpG clusters and there is an inverse relationship between CpG methylation and transcriptional activity. Therefore, quantitative methylation analyses have become an effective approach for cancer gene studies.
CpG island can be detected in DNA using a variety of assay techniques, including Southern blot analysis (SB), RE-PCR, bisulfite genomic sequencing (BGS), bisulfite modification followed by the polymerase chain reaction, and pyrosequencing.
